Gene
apdB
Protein
Cytochrome P450 monooxygenase apdB
Organism
Emericella nidulans (strain FGSC A4 / ATCC 38163 / CBS 112.46 / NRRL 194 / M139)
Length
498 amino acids
Function
Cytochrome P450 monooxygenase; part of the gene cluster that mediates the biosynthesis of aspyridones (PubMed:17369821). The polyketide-amino acid backbone is first assembled by the PKS-NRPS hybrid apdA (PubMed:17369821). ApdA catalyzes three Claisen condensations, as well as beta-keto processing and methylation (PubMed:17369821). Because apdA lacks a designated enoylreductase (ER) domain, the required activity is likely provided the enoyl reductase apdC (PubMed:17369821). Upon formation of the polyketide backbone on the thiotemplate, the triketide is transferred to the NRPS module and linked to tyrosine (PubMed:17369821). The presence of a C-terminal reductase domain suggests that the hybrid molecule is downloaded by reductive cleavage (PubMed:17369821). A number of oxidative steps are required for the transformation of the resulting pyrrolinone via a tetramic acid intermediate (PubMed:17369821). ApdB may have a role in the oxidation of the tetramic acid (PubMed:17369821). ApdB and ApdE might also be involved in the cryptic pyridone rearrangement (PubMed:17369821). Finally, the transformation of aspyridone A into aspyridone B is likely catalyzed by the predicted FAD-dependent monooxygenase apdD (PubMed:17369821).
