Function
Key regulator of TP53BP1 that plays a key role in the repair of double-strand DNA breaks (DSBs) in response to DNA damage: acts by promoting non-homologous end joining (NHEJ)-mediated repair of DSBs (PubMed:15342490, PubMed:28241136). In response to DNA damage, interacts with ATM-phosphorylated TP53BP1 (PubMed:23333306, PubMed:28241136). Interaction with TP53BP1 leads to dissociate the interaction between NUDT16L1/TIRR and TP53BP1, thereby unmasking the tandem Tudor-like domain of TP53BP1 and allowing recruitment to DNA DSBs (PubMed:28241136). Once recruited to DSBs, RIF1 and TP53BP1 act by promoting NHEJ-mediated repair of DSBs (PubMed:23333306). In the same time, RIF1 and TP53BP1 specifically counteract the function of BRCA1 by blocking DSBs resection via homologous recombination (HR) during G1 phase (PubMed:23333306). Also required for immunoglobulin class-switch recombination (CSR) during antibody genesis, a process that involves the generation of DNA DSBs (By similarity). Function
Negatively regulates telomere length by preventing telomere elongation or promoting degradation of the telomere ends. Recruited to telomeres by interaction with the C-terminus of RAP1, which binds directly to telomeric repeat DNA. This may create a negative feedback loop in which the addition of new telomere repeats creates binding sites for inhibitors of telomere length extension. May also influence the balance of transcriptional silencing at telomeres and the silent mating type locus HMR, which is mediated by SIR (Silent Information Regulator) proteins including SIR3 and SIR4. RIF1 competes with SIR proteins for binding to the C-terminus of RAP1. In the absence of RIF1, a limiting cellular pool of SIR proteins may preferentially associate with RAP1 at sub-telomeric loci, causing enhanced telomeric silencing and attenuated silencing of the HMR locus.

Regulates the timing of initiation of DNA replication (PubMed:29746464, PubMed:30277458, PubMed:26022086). Functions in copy number control by promoting the underreplication of DNA, which is found in many late replicating euchromatic regions of salivary gland polytene chromosomes (PubMed:30277458). Promotes underreplication by localizing to active DNA replication forks in a partially SuUR-dependent manner, and inhibiting replication fork progression (PubMed:30277458). Might also work as an adapter to recruit Pp1-87B to multiple sites on the chromosome and may function with Pp1-87B to mediate underreplication (PubMed:26022086, PubMed:29746464, PubMed:30277458). Plays an essential role in embryonic development, in the transition from larvae to pupae and, probably, in proliferating tissues later on (PubMed:26022086, PubMed:29746464). In embryos, during mid-blastula transition, binds to and selectively delays the replication of large blocks of repetitive DNA satellite sequences during S phase in response to the activity of Cdk1; maternal Rif1 is specifically required for the normal extension of S phase 14 (PubMed:29746464). Unlike mammalian orthologs, does not appear to play a role in DNA damage repair (PubMed:22712556, PubMed:26022086).
