Results
Activity Assay:
Contestant DNA was resuspended in water and 10 ng was used as template in a 25 µL PCR reaction with Q5 DNA polymerase. DNA was amplified using a standard cycling protocol and 3.3 µL was used as template for 33.3 µL cell-free reactions. Each reaction was diluted 100X and the TEV protease activity assay was carried out using the SensoLyte® 520 TEV Protease Assay Kit. Fluorescence at 520 nm was recorded at the 20 min time point.

Liberum is proud to announce the winner of this the Protein Design Games (Winter RosettaCon 2024) is Team 6. The team designed the three top-performing TEV protease sequences and will take home the cup, the bragging rights and the monetary award generously offered by the RosettaCommons.
