The first step of swainsonine biosynthesis is the condensation of one malonyl-CoA unit with one activated pipecolic acid residue by swnK (PubMed:28381497). The presence and positions in swnK of 2 reductase domains suggests that the intermediate released from this enzyme has a hydroxyl group (PubMed:28381497). This intermediate should cyclize to generate a C3=N(+) iminium ion that may be reduced by the action of one of the oxidoreductases swnR or swnN, leading to 1-hydroxyindolizidine (PubMed:28381497). Subsequent oxygenations must occur at carbons 2 and 8, probably via the activities of the dioxygenases swnH1 and swnH2 (PubMed:28381497). Furthermore, a second iminium ion intermediate, C9=N(+), generated during this step is also reduced by one of the oxidoreductases swnH1 or swnH2 to generate swainsonine (PubMed:28381497).
