Sequence

Gene
higB
Protein
mRNA interferase toxin HigB
Organism
Escherichia coli (strain K12)
Length
104 amino acids
Function
Toxic component of a type II toxin-antitoxin (TA) system. A probable translation-dependent mRNA interferase. Overexpression causes cessation of cell growth and inhibits cell proliferation via inhibition of translation; this blockage is overcome by subsequent expression of antitoxin HigA. Overexpression causes cleavage of a number of mRNAs in a translation-dependent fashion, suggesting this is an mRNA interferase. mRNA interferases play a role in bacterial persistence to antibiotics; overexpression of this protein induces persisters resistant to ciprofloxacin and ampicillin (PubMed:21788497).
Similarity
Belongs to the HigB mRNA interferase family.

Function
Toxic component of a type II toxin-antitoxin (TA) system (PubMed:19423702, PubMed:24257752, PubMed:8645296). A ribosome-associated translation-dependent mRNA interferase. Inhibits translation by sequence-specific cleavage of mRNA. Prefers either in-frame or out-of-frame 5'-AAA-3' codons (lysine). Cleaves also the first three AAAs of stretches of four or more A sequences. 20% of codons containing AA are cleaved and occassionally cuts even at a single A (PubMed:19423702).
